In April of 1951, readers who turned the pages of PNAS were treated to a surprise of multiple proportions: seven studies from the same authors, published back-to-back, all on the subject of protein structure (Pauling and Corey, 1951 a, b, c, d, e, f and g). The papers extended prior research published a month earlier in PNAS (Pauling et al., 1951) and another paper published a year earlier in the JACS (Pauling and Corey, 1950). An understanding of the basic structure of proteins provided the key to understanding as to how most of the proteins function in the cell at the molecular level. ‘Since Pauling and Corey’s famous series was first published, the ideas they expounded have proceeded from hypothesis to accepted fact to common knowledge’. Further verification of their work came in 1958, when it was shown that the threedimensional structure of myoglobin was composed almost solely of alpha helices (Kendrew et al, 1958). A few more notable pioneering works in this field include those of Linderstrøm-Lang (1952), Bell (1954), Sanger (1956) and Phillips (1965). Series of works on protein structure thus followed and elucidation of the structures of many complex proteins such as haemoglobin, tobacco mosaic virus have been established in the long run, along with the establishment of the Protein Data Bank (PDB) in 1971 at Brookhaven National Laboratory as a repository for threedimensional structural data of proteins and nucleic acids. When it was founded, the resource contained just seven structures. The PDB now headquartered at Rutgers University and funded by NIH houses more than 50,000 structures. |
