Description Leishmaniasis is the second most important protozoal disease after malaria and visceral leishmaniasis is the most fatal among all types of leishmaniasis. In present scenario about 12 million people are reported to be infected worldwide [WHO]. Till now there is no any effective vaccine against leishmaniasis. The toxicity, persistence of side-effects and the growing resistance of the clinically used drugs are still a matter of great concern. The search for antileishmanial agents has been exhaustive. Alternative drugs, such as amphotericin B and pentamidine, also have unpleasant side effects .So there is an urgent need for the development of alternative drugs.
Topoisomerases are essentially dualistic in nature, it can catalyze essential cellular reactions as well as it can also inflict great harm to the genome of an organism. Parasite Topoisomerases are sufficiently distinct from their human counterparts and this may allow differential chemical targeting and will therefore make good cellular targets. DNA topoisomerase I inhibitors have been widely studied in cancer therapy but despite being phylogenetically unique, having anomalous dimeric structure and its dual location –associated with both genomic DNA and kDNA , the trypanosomatid enzyme has not been fully exploited
Here we have evaluated Copper salycylaldoxime (CuSAL), a previously reported anticancer compound, as a potent chemotherapeutic drug against visceral leishmaniasis. We report that CuSAL inhibits DNA topoisomerase I of Leishmania donovani (LDTOP1LS) and causes apoptosis of the parasite. CuSAL has almost no any inhibitory effect on human topoisomerase I (HTOP1).
Recently topoisomerase inhibiting property of flavonoids have encouraged researchers to use its efficacy against leishmania parasites. Presence of flavonoids in Moringa oleifera, ginseng and Ginkgo biloba encouraged us to use its efficacy against the L.donovani parasite.
Here we have reported that the extracts from Moringa Oleifera, Ginko Biloba and ginseng show potent antileishmanial activity. All the active extracts of these plants were also able to potentially inhibit L.Donovani topoisomerse.
