Maydis leaf blight (MLB) caused by Bipolaris maydis is one of the most
important fungal diseases of maize. A study was conducted for grouping and
histopathological characterization of ten Indian maize inbred lines against MLB
disease. On the basis of phenotyping, they were categorized as five resistant inbreds
viz. SC-7-2-1-2-6-1, SC24 (92)-2-3-2-1-1, DMSC-16-2, V-351-1-4, HKI MBR-139-2
and five susceptible inbreds viz. CM-119, DMSC-28, HKI PC-8, HKI 1040-C2,
V341.The leaf hair density was high in resistant genotypes (299.33) and significantly
less in susceptible genotypes (61.00). Stomata count was found less (4.20) in resistant
genotypes as compared to susceptible genotypes (11.30). Scanning electron
microscopy (SEM) revealed more deposition of wax around the stomata of resistant
genotypes but less in the susceptible genotypes. A dense layer of mycelial mat of B.
maydis was visualized in susceptible genotypes than in resistant genotypes.
Combination of SEM and light microscopic studies revealed that B. maydis penetrated
maize leaves through stomata and colonized mesophyll cells. Transmission electron
microscopy (TEM) clearly showed the destructive effect of the fungal pathogen on the
mitochondria and other cellular organelles. Infra Red Gas Analyzer (IRGA) was used
to determine the behavior of physiological parameters of the host plants.
Photosynthetic rate was more in uninoculated (healthy) plant than in the inoculated
(diseased) plants. Under inoculated conditions photosynthetic rate was significantly
high in resistant lines than susceptible inbred lines. Overall photosynthetic rate was
gradually reduced with the advancement of time. The transpi ration rate (TR) of
inoculated plants was higher than the uninoculated plants at 5 DAI, whereas at 40 DAI,
it was decreased in both inoculated and uninoculated plants. Under inoculated
condition TR was higher in susceptible genotypes. Stomatal conductance (Cs)
decreases with the passage of time under inoculated condition. But a differential
behavior of Cs was found among the tested maize inbred lines under uninoculated
conditions
