Random Amplification of Polymorphic DNA (RAPD) was carried out to investigate
the genetic diversity and phylogenetic analysis in twenty Nostoc strains procured
from Culture Collection of Cyanobacteria at CCUBGA, IARI belonging to different
geographical origin. Twelve single primer and ten dual primer combinations were
used for the purpose. With both the single as well as dual primer combinations,
reproducible and completely polymorphic amplification profiles were obtained for
all strains. The RAPD analysis also produced strain specific unique bands which are
intended to be useful for identification of those strains. At least one to a maximum of
two unique bands were produced by different primers when used singly or in
combination. For sixteen strains out of twenty, strain specific bands were found to be
generated. Cluster analysis revealed a vast heterogeneity among these Nostoc strains
and no clustering based on geographical origin was found. It was also observed that
morphological data do not necessarily correspond to the genetic data in most of the
cases. The level of similarity of the strains ranged from 0.06 to 0.94. CCC92 (Nostoc
muscorum) and CCC48 (Nostoc punctiforme) showed a high degree of similarity in
all cases which was well supported by high bootstrap values. Fitness of the clusters
were good in most of the cases while the bootstrap values were not very high except
a few clusters. Cluster analysis based on HIP and RAPD primers used singly as well
as in combination showed a good fit.
