Using a widely grown and adapted wheat variety HD2932 as recurrent parent three near isogenic lines (NILs) carrying leaf rust resistance gene, Lr19, stem rust resistance gene, Sr26 and yellow rust resistance geneYr10 were developed just in a period of two and half years by raising two crops per year (main crop at IARI New Delhi and off season nursery at IARI regional station, Wellington, TN) with the help of marker assisted foreground and background selection. The results of background analysis indicated high genomic similarity of NILs with their recurrent parent which varied from 94.38 to 98.46%. The resistance to leaf rust, stem rust and yellow rust were stably expressed by the respective NILs as seedlings of each generation starting from F1 were tested using molecular markers linked to the resistance genes Lr19, Sr26 and Yr10. All the NILs were found to carry the targeted genes in the homozygous state in BC2F2 generations. The yield traits need to be recorded in BC2F3 generation before releasing as new varieties/essentially derived varieties. The potential applications of these NILs in gene introgression are discussed. |
