Leaf rust, (Puccinia triticiana Eriks. Syn. Puccinia recondita tritici Rob. Ex
Desm.) is one of the major diseases of wheat in India and all over world, causing
heavy yield losses. The breakdown of resistance due to evolution of new races reduces
the useful life of a cultivar. Development of cultivars with durable rust resistance is
one of the main objectives of breeders. Pyramiding of leaf rust resistance genes can
enhance the durability of rust resistance. Leaf rust resistance genes Lr19 and Lr28
were pyramided in the genetic background of high yielding cultivar HD2687. Near
isogenic lines of HD2687 carrying Lr19 and Lr28 were crossed to combine these two
genes. Marker assisted selection was practiced (MAS) in the F2 generation to identify
plants carrying both Lr19 and Lr28. SSR marker Xwmc221 and SCAR markers
SCS253 and SCS265 were used to detect Lr19 gene. SCS253 is linked with Lr19 in
repulsion phase and SCS265 in coupling phase. These two SCAR markers were used
in multiplex PCR reaction so as to identify plants homozygous and heterozygous for
Lr19. The results were further confirmed with co-dominant marker Xwmc221. Plants
carrying Lr28 were identified using SCAR marker SCS421. All the markers used
showed normal expected segregation in F2 generation. Phenotyping for leaf rust
resistance was done with race 77-5 at adult plant stage. F2 population segregated into
expected 15 resistant: 1 susceptible plant. Fifty one plants were identified to carry

Lr19 in homozygous state along with Lr28 in either homozygous or heterozygous
condition.
