Polymerase chain reaction (PCR) analysis was used to study the inheritance
of transgene in F2 and backcross population derived from the cross of homozygous
donor lines with Swarna. Chi-square analysis of goodness of fit revealed Mendelian
segregation in F2s of SGR2E1/Swarna and BC2F2s of SGR2R1/Swarna which
indicated that the transgene was inherited as a single dominant locus. Segregation
distortion was observed in F2s of cross derived from SGR2R1/Swarna when
analysed for both 3:1 and 1:2:1 ratio. The degree of distortion was different between
populations of crosses SGR2R1/Swarna and SGR2E1/ Swarna, and the distortion
was more distinct in SGR2E1 than SGR2R1 derived population. The observed high
degree of segregation distortion may be attributed to various factors as effect of
background genome, preferential transfer of transgene through male gamates and
presence of transgene in the chromosomal regions prone to display segregation
distortion.
