Description The aim of present study was to ascertain the efficacy of Farmer friendly Boar semen extender (FBSE) (50C), NBSE and PRIMXcell (15-170C) extenders in preserving crossbred Large White Yorkshire boar semen at varied intervals (1/2, 24, 48, 72 and 96 h of storage).
A total of 32 ejaculates (8 from each boar) from 4 boars were collected during the study. The mean ejaculate semen volume, colour, consistency, mass activity, concentration and total number of spermatozoa of ejaculate of crossbred LWY boar semen was 152.09 ± 12.22 ml, Grey white, thick milky, 4.34 ± 0.11, 444.31 ± 50.70 million / ml and 63.35 ± 9.03 billion /ejaculate. The individual motility, live spermatozoa, acrosomal integrity, HOST reacted spermatozoa and morphologically normal spermatozoa of the fresh semen was 84.06 ± 1.19, 85.10 ± 0.76, 97.25 ± 0.24, 67.68 ± 2.46 and 91.21 ± 0.81 per cent, respectively. The per cent of head, mid piece and tail abnormalities of fresh semen were 2.37 ± 0.26, 4.21 ± 0.63 and 2.00 ± 0.65, respectively. The mean time for MBRT and RRT was, 3.40 ± 0.25 and 6.68 ± 0.23 min, respectively, and the mean pH of the fresh semen was 7.38 ± 0.02. acrosomes, morphologically normal spermatozoa, head abnormalities, mid piece abnormalities, tail abnormalities and pH ranged from 79.21 ± 1.29 to 55.31 ± 2.29, 83.00 ± 0.83 to 66.34 ± 1.43, 52.28 ± 1.66 to 21.37 ± 0.77, 94.75 ± 0.26 to 85.06 ± 0.65, 87.53 ± 0.57 to 81.00 ± 0.75, 2.78 ± 0.18 to 5.71 ± 0.52, 6.18 ± 0.43 to 9.81 ± 0.67, 3.50 ± 0.53 to 3.59 ± 0.44 and 6.57± 0.08 to 6.74 ± 0.11 respectively in FBSE extender during 1/2h to 96h of storage. Mean individual motility, live spermatozoa, HOST reacted spermatozoa, intact acrosomes, morphologically normal spermatozoa, head abnormalities, mid piece abnormalities, tail abnormalities and pH ranged from 77.96 ± 1.41 to 44.84 ± 2.72, 79.46 ± 1.81 to 59.81 ± 2.12, 50.40 ± 2.04 to 19.43 ± 1.22, 95.34 ± 0.33 to 86.31 ± 0.45, 89.65 ± 0.61 to 83.78 ± 0.69, 2.93 ± 0.17 to 5.40 ± 0.38, 5.78 ± 0.45 to 7.81 ± 0.63, 1.65 ± 0.21 to 3.03 ± 0.49 and 6.81 ± 0.11 to 6.95 ± 0.11, respectively in NBSE extender during 1/2h to 96h of storage. Mean individual motility, live spermatozoa, HOST reacted spermatozoa, intact acrosomes, morphologically normal spermatozoa, head abnormalities, mid piece abnormalities, tail abnormalities and pH ranged from 66.40 ± 2.15 to 1.96 ± 0.72, 75.03 ± 1.84 to 49.53 ± 3.20, 36.31 ± 1.13 to 1.59 ± 0.80, 90.34 ± 0.52 to 67.15 ± 1.24, 89.90 ± 0.55 to 84.68 ± 0.76, 3.37 ± 0.33 to 4.96 ± 0.35, 4.34 ± 0.35 to 7.68 ± 0.50, 2.37 ± 0.32 to 2.71 ± 0.35 and 7.74 ± 0.09 to 8.23 ± 0.06, respectively in PRIMXcell extender during 1/2 h to 96 h of storage. In each and every phase of storage the per cent of individual motility, live spermatozoa, HOST reacted spermatozoa and Intact acrosomes were decreased in each extender, but FBSE and NBSE were significantly differed (p≤ 0.01) from PRIMXcell extender.
The extended semen was subjected to CASA and recorded various motility and velocity parameters. The motility parameters of CASA in FBSE and NBSE were significantly (p≤ 0.01) differed from PRIMXcell extender during storage of 1 h to 48 h. In all extenders, the total motility, progressive motility and rapid motility were decreased significantly (p≤ 0.01) with increased duration of storage i.e. Total motility was decreased from 90.45 ± 1.25 to 36.20 ± 1.84; 82.75 ± 1.51 to 40.15 ± 1.11 and 73.55 ± 2.57 to 15.10 ± 1.39, progressive motility from 75.35 ± 1.23 to 21.75 ± 1.22; 65.05 ± 2.14 to 29.35 ± 1.06 and 48.85 ± 2.11 to 3.50 ± 0.78 and rapid motility from 90.45 ± 1.25 to 36.20 ± 1.84; 82.75 ± 1.51 to 40.15 ± 1.11 and 73.55 ± 2.57 to 15.10 ± 1.39, respectively in semen extended with FBSE, NBSE and PRIMXcell extenders during the storage of 1h to 48 h. In FBSE, NBSE and PRIMXcell extenders, VAP (μm/sec) of spermatozoa was ranged from 57.34 ± 2.20 to 57.99 ± 2.08; 60.42 ± 1.91 to 54.66 ± 1.29 and 64.31 ± 2.51 to 58.98 ± 1.68, VSL (μm/sec) of spermatozoa was ranged from 47.49 ± 2.19 to 39.52 ± 1.52; 51.97 ± 1.88 to 37.26 ± 1.37 and 46.36 ± 2.18 to 39.84 ± 2.51; VCL (μm/sec) was significantly (p ≤ 0.01) increased from 100.59 ± 3.64 to 125.09 ± 3.51 in FBSE, 91.69 ± 7.51 to 135.97 ± 6.70 in NBSE and 122.20 ± 4.78 to 140.16 ± 6.47 in PRIMXcell (p ≤ 0.05) extenders, on 1h to 48 h of storage. The overall mean per cent of WOB was 49.98 ±1.13, 52.39 ± 2.18 and 48.98 ± 1.27; ALH (μm) was 7.04 ± 0.29, 7.47 ± 0.37 and 7.72 ± 0.45, Whereas BCF (Hz ) (p ≤ 0.05) was 37.52 ± 1.27, 34.67 ± 1.04 and 39.58 ± 1.28 in FBSE, NBSE and PRIMXcell extenders, respectively. However, the overall mean of STR was 72.95 ± 1.43, 75.06 ± 1.69 and 69.46 ±1.51; LIN was 37.53 ± 1.50, 41.68 ± 2.60 and 35.83 ± 1.54 per cent in FBSE, NBSE and PRIMXcell extenders, respectively.
Conventional and CASA parameters evaluation of extended semen revealed that both FBSE and NBSE extenders exhibited better keeping quality of extended semen. However, FBSE is the best option for preservation of boar semen under field conditions since it maintains the optimum motility, viability, acrosome and functional integrity of spermatozoa at 50C rather than at 15-180C on storage.
