Description Escherichia coli are a major causative agent of urinary tract infections (UTI) in companion animals
especially in dogs and cats. One hundred and three urine samples were collected and were cultured on
BA,MLA and EMB agar. Out of the 103, 25 were found positive for E. coli organisms. The positive E. coli
strains were subjected to antimicrobial susceptibility testing, PCR for the identification of antibiotic
resistance and virulence genes, sequencing and phylogenetic analysis. The isolates were resistant to
ampicillin, ciprofloxacin and tetracycline but sensitive to chloramphenicol, meropenem, gentamicin and
amikacin. The PCR assay detected blaAMPC (100%), blaTEM (60%), cmlA (16%), ParC (100%), GyrA
(100%), GyrB (4%), tet(A) (84%), tet(B) (52%) and tet(D) (12%) in the E. coli isolates as antibiotic
resistance genes, where aer (80%), pap (46%), sfa (48%), afa (32%), hly (20%) and cnf1 (20%) were
identified as virulence genes. None of the isolates harbor blaSHV, floR, tet (E), tet (D) and cnf2 genes.
Sequencing and phylogenetic grouping of the detected aer, afa, sfa, cnf1, hly and pap virulence genes
sequence revealed maximum nucleotide identity with several isolates retrieved from the GenBank data base,
and these hly, sfa, cnf1 and pap genes sequences matched to phylogenetic group B2, afa to phylogenetic A
and aer to B2 and D phylogenetic group. Cultural isolation of bacteria, antibiotic sensitiviy and molecular
characterization is considerd to be the gold standard method for the diagnosis of UTI in dogs and also aid in
selection of relevant antimicrobial threrapy.
