The present study evaluated the association between the SNPs in IL1 A 889 4845 IL1B 511 3954 and FCGR2A H131R FCGR2B Ile 232Thre TNFA 1031 863 and periodontitis susceptibility in a hospital based cohort from South India In addition a co relation of the gingival crevicular fluid levels of Interleukin1 beta and Tumor necrosis factor alpha with the SNP genotype IL1B 511 3954 TNFA 1031 863 was also performed to assess the functionality of the SNPs Linkage disequilibrium analysis and Multi factorial dimensionality reduction analysis was also performed to identify the presence of any interaction between SNPs of the genes evaluated A combination of Sanger sequencing and Real time SNP genotyping methods was used to identify the genotypes and cytometric bead array for the protein assessment in gingival crevicular fluid A significantly higher distribution of the variant alleles in IL1B 3954 IL1RN FCGR2A H131R was observed amongst subjects with chronic periodontitis An increase in risk OR ranging from 175 19 was observed when the subjects carried the variant alleles of the IL1B 3954 IL1RN FCGR2A H131R The carriage of CT variant of IL1B 3954 resulted in an increased production of Interleukin 1 beta protein The distribution of the variant alleles of the rest of the SNPs examined did not show any significant difference in the distribution between healthy controls and patients with chronic periodontitis MDR analysis revealed a significant gene interaction between the IL1B 511 3954 and FCGR2A H131R and FCGR2B Ile232Thre SNPs In a subgroup analysis of a 25 50 age cohort 352 participants a significant gene interaction was also observed between the IL1B 3954 and FCGR2A H131R These two SNPs occurred together in patients with periodontitis with a frequency that was more than observed by chance Haplotype analysis for the IL1A IL1B SNPs revealed TGTT was associated with increased periodontitis risk and CTCC was of a protective nature Haplotypes were not formed for the remaining SNPs examined.
