newline ABSTRACT newlineThe present research investigate that Pergularia daemia Forsk is pharmacologically use as analgesic, anti-inflammatory, antipyretic, antioxidant activity. The plant also shows anthelmintic, laxative, property. Use in urinary discharges, leucoderma, strangury, uterine complaints, facilitates parturition. The analgesic, anti-inflammatory and antioxidant activity of leaf and stem is still not reported, hence, leaf and stem were screened for its analgesic, anti-inflammatory and antioxidant activity. Microscopic study of leaves shows crystal distribution, stellate trichomes, crystals, actinocytic stomata, and star shaped trichomes. Stem shows two patches of meta- xylem opposite to each other. Petroleum ether extract of leaves and stem shows sterol, triterpenes and methanol extracts shows alkaloid, glycoside, tannins and flavonoids. Qualitative analysis was done by TLC and quantification by HPTLC. Active constituent separated by column chromatography and analyzed by IR, NMR, GC-MS.These extracts were screened for their pharmacological activities. Acute toxicity study was performed and dose of extract was calculated as 100mg/kg and 300mg/kg for both extract and for isolated component lupeol dose was 5-10 mg/kg. Activity was performed on extract as well as isolated component as analgesic by hot plate method and tail immersion method, anti-inflammatory by Carrageenan induce and cotton pellate granuloma method. Antioxidant activity was performed by DPPH and Nitrous oxide induce scavenging radical method. Petroleum ether and methanol extract of leaf and stem of Pergularia daemia show equipotent activity but petroleum ether extract of stem shows potent activity as compare to other hence further analysis were performed on total petroleum ether extract of stem. Phytoconstituents such as betasitosterol, lupeol and phytol were isolated from petroleum ether extract where as gallic acid and ellagic acid from methanol extract of stem. newlineQuantification of isolated compounds from both parts was performed by HPTLC — |
