Abstract Isolation of pure and high molecular weight DNA is essential for the analysis of complex genomes using molecular biology techniques. The protocol for the isolation of genomic DNA consist of two parts: a technique to gently lyse the cells and solubilise the DNA, followed by one of several basic enzymatic or chemical methods to remove contaminating proteins, RNA and other macromolecules.
