Immunity against the bovine intracellular protozoan parasite Theileria parva has been shown to be mediated through the destruction of parasite infected lymphocytes by MHC class I restricted CD8+ CTL. Six parasite proteins targeted by CTL from T. parva immune cattle have been identified and raised the prospect of a subunit vaccine. In this study, we identified nine CTL antigenic peptides on these six proteins with minimal peptide lengths of between 9 and 11 amino acids long. The bovine MHC (BoLA) class I alleles that restrict these epitopes were identified by functional screening of BoLA class I cDNA clones and showed that individual alleles restricted five of these peptides and two additional alleles were each found to restrict two peptides. These data were used to evaluate the ability of available bioinformatics software to successfully identify bovine CTL epitopes. A high degree of agreement was observed between the positions of epitopes predicted to bind human or mouse MHC class I molecules and those actually recognized by bovine T cells. These results support the application of available software to assist in the screening of CD8+ T cell epitopes recognised by veterinary Species, although the efficiency of this process will undoubtedly be improved as more Species-specific MHC-binding motifs are developed.
