Description To determine the mean lethal concentration (LC50), a series of experiments were performed. For each isolate involved, 5 different concentrations were used (109, 108, 107, 106 and 105 granules/ml) with 6 replicates per concentration. Besides, one control was used for each series of experiments.
A stock solution with a concentration of 109 granules/ml was prepared. It was diluted by factor 4 (3ADE:1stock dilution). From this, four dilutions were made successively using factor 9 (8 ADE:1 from the previous dilution).

For viral inoculation, filter paper discs with 800 to 1000 eggs were immersed in the viral solution. These filter paper discs were allowed to dry at room temperature, and placed in petri dishes, then incubated at 24°C until larvae hatched. For control, they were immersed in 1 ml of distilled water.

After larvae hatching, 50 neonatal larvae, previously inoculated, were placed in approximately 100g of potatoes in ½ liter plastic containers. Larvae were incubated in a growth chamber at 24°C. Then, two evaluations were carried out (At 14 and 21 days after the inoculation). In the evaluations the number of larvae with symptoms of a PoGV infection (white color, slow movement and unable to pupate) and the number of pupae was recorded.
