The present investigation generates a rapid and convenient stable expression protocol based on vacuum infiltration of Agrobacterium tumefaciens cells into moth bean cotyledonary nodes. Vacuum infiltrated tissues showed transient GUS expression than did non infiltrated tissues. The leaflets arising from the transformed shoots on kanamycin supplemented media were found GUS positive during histochemical assay. We believe this to be the first report of this technique for putative gene expression in mothbean cotyledonary nodes. Mothbean cotyledonary nodes are one of the most regenerative tissues in legumes.
