Studies were undertaken to determine the phosphorus, protein and DNA contents of viable buffalo oocytes before and after in vitro maturation with supplementation of different concentrations of IGF-I (0, 25, 100, 200 ng/ml) in culture medium. Oocytes were aspirated from abattoir derived buffalo ovaries. The oocytes with homogenous ooplasm and compact cumulus cell layers were preliminarily selected and stained with trypan blue to test their vitality. The concentration of 200 ng/ml of IGF-I in the maturation medium was optimum for in vitro maturation of buffalo oocytes. IGF-I stimulated in vitro oocyte maturation in a dose-dependent manner. The phosphorus, protein and DNA were significantly higher in oocytes after culture compared to those before culture in all treatments. Not Available |
