A method of microbiological assay with an additional trienzyme extraction method in the order of protease, α-amylase and finally conjugase along with the standard microbiological assay using Lactobacillus rhamnosus as an assay organism in casein based media for the estimation of folate was evaluated for its applicability in milks from different Indian milk species. The estimated average values of folate in milk from Indian milk producing animals such as goat, cow, sheep and buffalo were found to be 10 µgL-1, 44 µg L-1, 56 µg L-1 and 60 µg L-1, respectively. Buffalo milk was found to be the richest source of folate among the milks analyzed. The performance of this method has been evaluated by comparing the values of folate level with those available in literature. The folate estimation by this method, though a little labor intensive has been found to be highly selective, sensitive, reproducible, and gives the total folate content of the sample analyzed.
