Soil samples were collected from the mango orchards at CISH, Rehmankhera for isolation of EPNs. A population of Steinernema sp. was extracted from soils by Galleria mellonella baiting technique (Bedding and Akhurst, 1975). The samples were purified for Koch postulates and maintained in the BOD incubator at 15 C. For molecular studies, DNA was isolated from fresh infective juveniles (one week old) by using 50 µl of worm lysis buffer (50 mM KCl, 10mm tris, 205 mm MgCl2, 0.45% NP40, 0.45% Tween 20, 0.01% gelatin and 60 µg/ml proteinase K) and homogenizing the nematodes. The suspension was incubated at 95 C for 10 min, 65 C for 1 h, and at-80 C for 2 h and finally stored in-20 C for further use. DNA was checked for its quality and quantified using UV-spectrophotometry.
