Description A study was conducted to manage diamondback moth (Plutella xylostella), an insect pest of cabbage crop, by applying "a biopesticide Bacillus thuringiensis Bed subsp kurstaki (Etk) at New Delhi. A simple method is presented for the estimation ofcrude (total) protein as well as soluble (cry) proteins in two Bacillus thuringiensis subsp kurstaki formulations, i e HIL-Btk and Biolep. The total protein content was found to be 4.76% for HIL-Btkand 5..9% for Biolep. The soluble (cry) protein was 3.53% for HIL-Btk and 4.3% for Biolep. Perfonning SDS-PAGE complemented the protein estim1tion. Bioassay studies were also carried out to test efficacy of the 2 formulations against Plutella xylostella in vivo (field) as well as in vitro.. LCso was found to be 0.0120.014 % for the two Bacillus thuringiensis subsp kurstaki formulations. The methodology developed in this paper was tested for another two commercial formulations ofBtknamely Delfin (WG) procured from Sandoz and Wockard from Halt India Ltd available in India. These also showed the presence ofa band in SDS-PAGE at 134 KD, confinning their ability to manage lepidopetam pests. The nlethod described in the present investigation for the estimation of protein content, can be used for estimation of active ingredient in tenns of total and soluble protein in Bacillus thuringiensis subsp kurstaki based formulations available in the market. The estimation ofactive ingredient ofBacillus thuringiensis subsp kurstaki fonnulations is essential for its quality control. The method presented in the paper for estimation of Bacillus thuringiensis subsp kurstaki protein is useful as a routine laboratory method as it is simple and can be easily adopted. Studies also revealed that bioassay method or SDS-PAGE must be complemented for the estimation ofprotein in order to compare the efficacy ofdifferent fonnulations.
