This study was conducted to overcome the consequences of Pseudomonas fluorescens (PF) infections in fishes
by explore the possibility of application of lytic bacteriophages against its antibiotic resistant isolates. Isolation of thirty eight
lytic P. fluorescens phages (designated as PFP1 – PFP38) specific to twenty isolates of PF was made from the water and bottom
sediments of various geo-climatic regions of North India. Occurrence of varied size plaques ranging between 4.15 to 11.70 mm
during 18 – 24h incubation in overlaying method was recorded in the samples of fishpond, sewage canal and Nainital Lake. Two
lytic P. fluorescens phages (PFP1 and PFP12) exhibited broader host range as they infected 11 of 20 isolates of P. fluorescens and
considered as polyvalent. They were made of hexagonal head of 35nm and long non contractile tail of 70nm and belonged to
family Siphoviridae, a member of ds DNA virus. The mean log10 cfu value of virulent P. fluorescens isolate (PF4) in 6 - 8h old
broth culture post inoculated (PI) with polyvalent phages ( PFP1 and PFP12) at 0.01 MOI individually or in combinatorial
treatment got reduced by 2 logs at 24h. The mean OD values of broth culture dropped to 0.49 from 1.0 and phage titers accrued
(pfu value from 108 to 1010 ) concurrently. The cocktail of these 2 polyvalent phages (in the ratio of 1:1 with 106 to 108pfu ml-1)
eliminated virulent P. fluorescens isolate (PF4) completely in its exponential phase in broth culture. It’s highly in vitro lytic
attributes explicates that it would a candidate species to combat with maladies caused by virulent P. fluorescens isolate as it did
not develop resistance to both the phages during entire period of our experimentation.
