Rajapuri Bale (AAB) is a popular cultivar of banana grown in northern parts of Karnataka especially in Bagalkote, Vijaypur and Belgaum districts. The demand for planting material of this cultivar is high and tissue cultured plantlets are not available because of the inherent problem in the initial establishment of the culture. The present investigation was carried out to optimize protocol for establishment of aseptic culture for facilitating large scale multiplication. Different types and levels of cytokinins viz., benzylaminopurine (BAP), thidiazuran (TDZ) and varying sizes of male flower explants were tested. Among the different cytokinin treatments, Murashige and Skoog (MS) basal medium enriched with 5 mg/l BAP along with 1 mg/l α-naphthalene acetic acid (NAA) and 150 ml/l coconut water recorded the maximum establishment of aseptic cultures (100.00%), low browning (+) and maximum greening and bulging (+++). Among different explant size tested, significantly maximum establishment of aseptic culture (100%), low browning (+) and maximum greening and bulging (+++) were observed with male flower explant size of 3 cm. |
